Water-soluble BODIPY and aza-BODIPY dyes: synthetic progress and applications

摘要: In recent years, boron-dipyrromethene (BODIPY) and boron-azadipyrromethene (aza-BODIPY) dyes have attracted considerable multidisciplinary attention due to their diverse applications. By introducing various hydrophilic groups, such as quaternary ammonium, sulfonate or oligo-ethyleneglycol moieties into the BODIPY core, the solubilities of these dyes in aqueous solution can be greatly improved while maintaining their high fluorescence quantum yields. Accordingly, applying these fluorescent dyes in aqueous systems to areas such as chemosensors, biomacromolecule labeling, bio-imaging and photodynamic therapy has been achieved. In this article, the recent progress on the synthesis, optical properties and application of water-soluble BODIPY dyes and aza-BODIPY dyes is reviewed.

文章从合成橡胶的化学基础出发,叙述了合成橡胶工业二十世纪以来的过去、现状,并展望未来。着重讨论了合成化学基础及相应聚合工艺过程的进步这两方面的问题。指出合成橡胶化学基础中活性聚合、可控聚合,包括茂金属催化剂仍为近期的重点;气相聚合工艺将会得到较大的发展;高效聚合釜的研究仍是核心问题;环保问题应更受到重视。

Genome reprogramming for synthetic biology

摘要: The ability to go from a digitized DNA sequence to a predictable biological function is central to synthetic biology. Genome engineering tools facilitate rewriting and implementation of engineered DNA sequences. Recent development of new programmable tools to reengineer genomes has spurred myriad advances in synthetic biology. Tools such as clustered regularly interspace short palindromic repeats enable RNA-guided rational redesign of organisms and implementation of synthetic gene systems. New directed evolution methods generate organisms with radically restructured genomes. These restructured organisms have useful new phenotypes for biotechnology, such as bacteriophage resistance and increased genetic stability. Advanced DNA synthesis and assembly methods have also enabled the construction of fully synthetic organisms, such as J. Craig Venter Institute (JCVI)-syn 3.0. Here we summarize the recent advances in programmable genome engineering tools.

Targeting “undruggable” c-Myc protein by synthetic lethality

摘要: Synthetic lethal screening, which exploits the combination of mutations that result in cell death, is a promising method for identifying novel drug targets. This method provides a new avenue for targeting “undruggable” proteins, such as c-Myc. Here, we revisit current methods used to target c-Myc and discuss the important functional nodes related to c-Myc in non-oncogene addicted network, whose inhibition may cause a catastrophe for tumor cell destiny but not for normal cells. We further discuss strategies to identify these functional nodes in the context of synthetic lethality. We review the progress and shortcomings of this research field and look forward to opportunities offered by synthetic lethal screening to treat tumors potently.

摘要: The advent of synthetic biology has ushered in new applications of cell-free transcription-translation systems. These cell-free systems are reconstituted using cellular proteins, and are amenable to modular control of their composition. Here, we discuss the historical advancement of cell-free systems, as well as their new applications in the rapid design of synthetic genetic circuits and components, directed evolution of biomolecules, diagnosis of infectious diseases, and synthesis of vaccines. Finally, we present our vision on the future direction of cell-free synthetic biology.

Flexural behavior of ECC hollow beams incorporating different synthetic fibers

摘要: Twelve ECC beams with three different fiber types, along with four normal concrete beams, were tested in this study to evaluate the influence of cross-sectional hollowing on their flexural performance. The fiber types used were nylon monofilament (NM), low-cost untreated polyvinyl alcohol (PVA), and polypropylene (PP). Three different square hole sizes of 60, 80, and 100 mm with cross-sectional hollowing ratios of 0.16, 0.28, and 0.44, respectively, were adopted for each group of beams in addition to a solid beam. All beams were tested under four-point loading using a displacement-controlled testing machine. The test results showed that ECC beams can mostly withstand higher cracking and ultimate loads compared to their corresponding normal concrete versions. The results also showed that both the ductility and toughness of the ECC beams are higher than those of the normal concrete beams and that the ductility values of the hollow beams with a hole size of 60 mm are higher than those of the corresponding solid beams. Moreover, hollow ECC beams with hole sizes of 60 and 80 mm exhibited a higher ductility than a solid normal concrete beam.

Study on a green synthetic method of TRPPMCl (M= Fe, Mn, Co)

摘要: A green synthetic method using mixted solvents of di-methyl formamide(DMF) and acids instead of single DMF to synthesize metalloporphyrins(TRPPMCl) from TRPPH and MCl metallization was proposed in this paper. A series of TRPPMCl (M= Fe, Mn, Co) were synthesized through this innovative synthetic method, and these complexes were characterized with various spectroscopic techniques, including IR and UV-Vis spectroscopy. The results showed that the metallization time was markedly reduced from about 10 h to 0.5 h in contrast with the conventional synthetic methods.

摘要: A synthetic mixture of real waste packaging plastics representative of the residue from a material recovery facility (plasmix) was submitted to thermal and catalytic pyrolysis. Preliminary thermogravimetry experiments coupled with Fourier transform infrared spectroscopy were performed to evaluate the effects of the catalysts on the polymers’ degradation temperatures and to determine the main compounds produced during pyrolysis. The thermal and catalytic experiments were conducted at 370°C, 450°C and 650°C using a bench scale reactor. The oil, gas, and char yields were analyzed and the compositions of the reaction products were compared. The primary aim of this study was to understand the effects of zeolitic hydrogen ultra stable zeolite Y (HUSY) and hydrogen zeolite socony mobil-5 (HZSM5) catalysts with high silica content on the pyrolysis process and the products’ quality. Thermogravimetry showed that HUSY significantly reduces the degradation temperature of all the polymers—particularly the polyolefines. HZSM5 had a significant effect on the degradation of polyethylene due to its smaller pore size. Mass balance showed that oil is always the main product of pyrolysis, regardless of the process conditions. However, all pyrolysis runs performed at 370°C were incomplete. The use of either zeolites resulted in a decrease in the heavy oil fraction and the prevention of wax formation. HUSY has the best performance in terms of the total monoaromatic yield (29 wt-% at 450°C), while HZSM5 promoted the production of gases (41 wt-% at 650°C). Plasmix is a potential input material for pyrolysis that is positively affected by the presence of the two tested zeolites. A more effective separation of polyethylene terephthalate during the selection process could lead to higher quality pyrolysis products.

Genomics and genetic breeding in aquatic animals: progress and prospects

摘要: Genomics focuses on dissection of genome structure and function to provide a molecular basis for understanding the genetic background. In a pivotal step, the expense of whole genome sequencing has been largely eliminated by the rapid updating of sequencing technology, leading to increasing numbers of decoded genomes of aquatic organisms, driving the aquaculture industry into the genomic era. Multiple aquatic areas have been influenced by these findings, such as accelerated generation shift in the seed industry and the process of breeding improved lines. In this article, we have summarized the latest domestic and international progress of aquatic animals in nine aspects, including WGS and fine mapping, construction of high density genetic/physical maps, trait-related marker/genes screening, as well as sex control, genome editing, and other molecular breeding technologies. Finally, the existing problems in this field have been discussed and five future counter measures have been proposed accordingly.
